Corrado,

we have been using cation exchange on a gram scale preparative scale for
our synthetic peptides for more than ten years and would be horrified to
have to spend this sort of money on a column.

Consider buying yourself a good moderate pressure empty column (we like the
Pharmacia XK series) and pack it with either Pharmacia S Sepharose HP or
BioRad MacroPrepS, we are using the latter at the moment because of its
availability in serious bulk quantities. I expect the total cost will be
less than $2000 for a setup big enough to bind many grams of recombinant
protein, if you choose a large enough diameter column (ie short and fat)
you will be able to achieve flow rates in the 20 to 50 ml/minute range. The
great thing about this approach is that, when you have pumped so much
garbage on the column that the top is discoloured and you're a bit
concerned about the chromatographic resolution, just open it up, suck off
the discoloured material and off you go again.

Save your money!
