I was relatively successful raising the temperature to 70C (column with
jacket) and dissolving my hydrophobic membrane peptide in the mixture of
trifluoroethanol/water (5/1), or even TFA alone. Mobile phase was ACN
water (1% TFA) with C4 column. I noticed that part of the loaded
material was washed from the column with a dead volume. Imagine
insoluble greasy peptide that is not going inside the pores at all. I
mean, check your very first eluted fractions - it may be there, not on a
column.
