Dear Joel,

There is a procedure for fatty acid acylation in Methods in Molecular
Biology, Vol 35: Peptide Synthesis Protocols Edited by M.W. Pennington and
B.M. Dunn 1994 Humana Press Inc.,Totowa, NJ Chapter 8 page 176.

They suggest 4 eq of fatty acid in 10ml DCM or DMF/gram of N terminal
deblocked resin. Activate fatty acid with 4.4 eq diisopropylcarbodiimide
and 8 eq HoBt and react overnight at 35 C with resin. Ninyhydrin
monitoring may indicate the need to double couple. After completion wash
3X with DCM,2X with DMF, 2X DCM and finally two methanol washes. The
peptide may require acetic acid or TFA for solubilization. MS analysis or
GC lipid analysis will confirm the fatty acid has coupled to the peptide.

I have never tried this protocol but years ago coupled palmityl chloride to
a peptide resin. Hope this helps.
