Len is correct that citraconylated lysine is not completly stable. I
recall that we added some additional citraconic anhydride just before
trypsin addition and some more citraconic anhydride was added during the
digestion. This of course inactivated some of the trypsin, but the
substrate protein remained protected. If lysine does not have to be
recovered, succinic anhydride can be used.
