Psyn: synthesis strategy and relative cost

Hi again and thank you for the answers of my previous message(s)!

I would like to know what other peptide synthesis facility would do if
they were in my situation: a researcher of our university wanted that I
synthesize a peptide (23 mer) that I will call xyz, and wanted some
modifications:

xyz
xyz-CYS
biotin-xyz
biotin -xyz-CYS

(so same peptide but with amino or C-terminus modified= 4 different products)

The C-terminus of the peptide xyz is arginine. He wanted the products 95%
pure and of course wanted the best price.

Would you synthesize it in one step using two resins (preloaded Cys Wang
resin, add Arg and then add Arg preloaded 2 Cltrytil resin) and take
aliquotes at the end for biotinylation? Or would you synthesize it more
than one time or else?

Would you charge for 1,2, 3 or 4 synthesis?

(in fact, I already did the synthesis in one step and charged for one
synthesis + 4 purification but the yield of one of the 4 products is not
enough (5 mg instead of 20) and he wants that I synthesize the peptide
again (free of charge). Before I do anything else for him, I would like to
know if my strategy was right and if my "relative" charge (price) was good
in comparison with other services. I know that the yield would not have
been a problem if I had synthesized them separately.
