David: The reagents used in the ABI C-terminal sequencer are formulated
to work with PVDF, the most widely used blotting material. Teflon is the
sample support that has to be employed in the HP C-terminal instrument
since the chemicals used in this sequencing method attack PVDF.

Siv: I would talk to ABI to help you to identfy a lab that runs
PVDF-blots unless you are willing to change your blotting method.
In that case, Ken Graham at City of Hope would be the guy to contact.
At ABRF he has shown spectacular runs with up to 12 residues identified
by substantially improving the original Bailey chemistry. If you are
happy with one or two interpretable cycles, the current HP 241 sequencer
will do the job.

PVDF has been reported to dissolve in some of the C-terminal sequence chemicals.
There are lit. on using LD telfon tape as a support material.

Is it possible to do C-terminal sequencing from a PVDF blot and
which facility can do this? How much material is needed for
C-terminal sequncing? We know the sequence and only need a few
