With regard to isozyme issue, we have single point mutants that show
similar shifts in MAPK kinase. If its an expressed protein, I've seen an
example where misincorporation occurs at arginine with high frequency that
cause a similar shift. I've also seen a case where a protein cyclized N-
to C-terminus during storage and it showed a shift. You may want to
consider lipid modification as well, if its partially membrane associated.
