Dear Els Van Geldre: I think your success might be greater in a different
forum, however, one can use a number of promoters that are available for
leaf-specific proteins. The dominant ones are for Rubisco, LHCP etc these
proteins are specific for the chloroplasts. The promoters should be
available from most labs that perform plant transformation, eg at
Wageningen or even in several labs in Belgium. The sequences for many of
the promoters are published so you should be able to PCR them out from
genomic DNA. I believe you will have better success if you directly
contact a local lab that has published studies on plant transformation
using one of these promoters. If you experience difficulties, please let
me know and I will put you in contact with people in my University and
Brazil who are currently using these constructs. Gautam
