Mike,
I was also interested in using autolytic peptides from LysC as internal
calibrants. However, the enzyme contains only 5 Lys residues, and the
theoretical fragments (not taking possible SS-bridgs into account) would =
be:
1-30 3090.5
31-49 1919.9
50-106 5911.6
107-155 5499.7
156-203 4872.3
204-268 6509.1

To check this in practical terms, I had 0.5 =B5g of LysC in Tris overnigh=
t
and analyzed the autolytic products by MALDI and found

1611.3
1748.3
2160.4
2246.4

(At that time I didn't "look" above m/z 3500). I have no idea what miscu=
ts
are behind these values!
