hi david,

the idea to look trough the novabiochem catalog is good, they do offer a
lot of nice AA for your needs. I worked a lot in the field of orthogonal
modifications of peptides, especially the NH2-side-chain of lys which was
usually bound to the resin. I have to say that I never tryed the
Alloc-group but Mtt and Dde worked out very well.
to deprotect Mtt you have to use 1 % TFA and 5 % TIPS for I think 30 min,
but I prefered three times 20 min which worked much better. with my
experience 2 % TFA were ok also and did not affect other side chains as
tBu and Boc neither the binding of the peptide to HMP-Resin.
though in some cases I had problems to remove all the Mtt groups in the
peptide (sometimes up to five of them) I tryed the Dde group which was
superior and deprotection with 2 % hydrazin never failed. if you need the
literature just send me an e-mail but usually the literature in the
nova-catalog is fine.
