Dear Ken,
I tell my collaborators to crosslink or covalently bind the Ab. So far we
used successfully CNBr-activated sepharose to couple the Ab as well as Ab
bound to protein A sepharose followed by DMP crosslink. Another option
would be to try the Affigel from Biorad. This brings the levels of Ab
normally down to femto-picomole. That means you will only detect it in the
heavy and light chain region. I analyzed once a non crosslinkt Ab-IP with
distinct bands besides the huge blobs for light and heavy chain. The bands,
independently of their respective MWT all gave Ab sequences with
NanoESI-MSMS.
