I apologize for making this into two submissions. In my haste to dash off a
quick note, I failed to clearly state my needs. We've made a peptide with a
phosphorylated Tyr using the standard Fmoc-Tyr(PO3H2)-OH and/or
Fmoc-Tyr[PO(OBzl)]-OH. This peptide showed great biological activity, but the
peptide appears to be dephosphorylated rather quickly. Many people have come up
with analogs of pTyr that are not so easily hydrolyzed. One example suitable for
Fmoc synthesis is Fmoc-Tyr(malonyl-di-OtBu)-OH sold by Bachem ($500/250mg).
I have checked all of the major peptide companies and I can't find a similar
compound for sale (even though I know of at least 2 others in the literature). I
was hoping that someone knew of a new product or new company that could help me.
I think I have my answer (which is that these compounds are not readily
available), but I didn't want you to think that I was just to lazy to scour the
catalogs to come up with something.
For the hard-core chemists lurking out there, has anybody made these compounds?
I guess I'd settle for a good procedure and hope for a student to come along and
synthesize this thing for me.
