If what you would like to calculate is the difference in the conformational
stability ( the deltaG of the folding) between the WT and the mutant
protein, a very good explanation in how to do it can be found in:

Creighton TE (ed): Protein Structure: A practical approach. Oxford, IRL
Press 1987, pp311-330.
Perhaps there is also a more recent edition.
