Dear Satya,
In case your glycopeptide is attached to resin via ester bond and/or if the
sequence contains Asp(OR), Glu(OR), there is a serious danger of
hydrazinolysis of these functions. In fact, they are chemically very
similar to OAc you are going to remove from Gal moiety.
I would recommend to carry out de-acetylation in solution after global
deprotection and cleavage from the resin.

Hello all, We are attempting to remove the -o-acetyl groups from the
galactose residue of a glycopeptide that has been synthesized in our
laboratory. We plan to deacetylate on solid support by addition of a
solution of hydrazine hydrate in methnol for 6 hr. Is there any other safe
method for o-acetyl removal from glycosyl residues. Any precaution to
take during such a cleavage?
