Re: protocol to deFmoc and cleave a fully protected peptide which is
off the resin and has
been precipitated with ether.
1). Add enough 20% piperidine/DMF to dissolve the peptide.
2). Let stir for 30 minutes.
3). Add ether to precipitate the 'defmoc'-peptide.
4). Wash precipitate with ether, three to four times.
5). Proceed to cleavage by adding the appropriate cocktail.
