Hi Harold--
This sort of thing happened to me when I messed up
the filtration step. I failed to notice that the
columns had not pre-spun, and as a consequence a
lot of buffer was dried down with the sample. On
the gel, the lanes completely criss-crossed each
other. Perhaps some of your samples have salt or
buffer in them?
