Hello ABRF'ers,
We have severe background problems with running "blanc" 2D gels.
2D-system : 1. sample prep : in-gel rehydratation using IPG strips 3-10
from Pharmacia
2. Laemmli-gel
Silver staining of a blanc 2D-gels reveals the presence of different spots
in the high MW region (>45kDa). The position of the spots in different
"blanc" gels is similar (some are real "spots", others looks like a train
of spots). These spots are also not reactive with "PAN"-anti-cytokeratin
antibody (broad spectrum antibody for keratin).
Spots are not CBB-R stainable but are well visible with silver stain.
Can anyone give some comments on this phenomenon?
Thanks in advance
Jos
