Jim,
Due to severe power outage resulting from an ice storm in the Washington DC
area, could not reply earlier. Hope you already have a solution. Still an
alternative one from our lab.
Put the excized gel piece between two plastic sheets. Roll over a glass
bottle applying some pressure. The gel will be mashed into hundreds of
microscopic pieces. Open the sandwich and collect with plastic spatula -
the edge of a .5 mm teflon spacer works very well (at this point you may
have to add about 10 mcl buffer.)
The pieces are so tiny that after digestion we routinely spin the mixture
through .45 mcm filtration unit (millipore UFC30HV25) - this also
squeezes out liquid from the gel.
