HI,

we have the upgrade since some time, and in principle it did work in our
hands. We load the wells one-by-one, and under these conditions the versi=
on
3.x tracker works OK for us. However, we detected a significant loss of
read length on our 96-gels compared to other gels. I have no exact idea w=
hy
there is this reduction, but I asume that the lanes are to narrow to buil=
d
up a real "band" in the gel, and the result is "blobs" of signal running
accross the detection zone. In addition, even the best tracking has troub=
le
in finding the centre of the lane in terms of data points on the gel imag=
e,
and also this may contribute the problem.

In practice, we now use 64-lane gels with the step-plates. The comb used =
is
the one from the 373 PE/Abi equipment. The result is 0.4 mm "thick" wells
which are realy easy to load. We reach good read length, and the gel
performance is OK.

One other point: During trying the PE/Abi 1000+ protocols, we very often
have spacing values which are too high for the automatic analysis softwar=
e.
This results in a "red" spacing value "9.00". It's due to the fact that t=
he
(sorry - stupid!!) software can not handle spacing values higher than 16
(why does PE/Abi recommend protocols which result in data which their own
software can not work with??). Has anybody a solution for this problem?
(beside setting the spacing value by hand to 17.00)
