In the course of developing a centrifugal system for desalting and
concentrating multiple samples in parallel from proteolytic digests, we had
occasion to investigate some problems associated with the use of reverse
phase materials. We've used 3 micron Vydac and 5 micron Zorbax C18, and
R2-10 Poros. For comparing our method, geloader tips and the ziptips, were
used. I hasten to add that while we've only used the ziptips a couple of
times, our general observations on the use of C18 materials for desalting
indicated that one must take great precaution to prevent the resin from
becoming dry during the procedure. Otherwise, it may not wet well enough
with the low organic required to wash away salts, detergents, or whatever
to achieve adequate ESI signals. If you are interested, we will be
presenting a poster on our observations in North Carolina and would like to
hear from others who have their own anecdotal experience.
