Dear Dr. Lyons,
We have used the EDGE (formerly AGCT) 96 - well miniprep system to do over
4000 minipreps for a small EST project. You need a centrifuge that has buckets
for "tall" microtiter plates, otherwise everything is standard. Using the
modifications as described in Utterback, T.R., McDonald, L.A., and Fuldner, R.A.
1995, "A reliable, efficient protocol for 96-well plasmid DNA miniprep with
rapid DNA quantification for high-throughput automated DNA sequencing", Genome
Sci. Technol. 1: 1-8, we have had consistently good sequencing results from
plasmids isolated from this system. The cost is about $1 per miniprep which is
cheaper than Qiagen (I have used both). Contact EDGE in Gaitherburg, MD at 301

I have a client who wants to submit 500 clones for sequencing,
and would appreciate not having to (a) run manual minipreps
on all of them, or (b) buy a robot or other expensive apparatus
for this one chore. The clones are currently in five 96-well
plates, presumably as bacterial stocks.

Does anyone have suggestions on how they should proceed? Any
experience with 96-well miniprep methods that would give good,
sequencable template and yet not cost too much? Source for
protocols?
