Again big peaks in the pth chromatogram,

First I thank all the ideas about my big peaks in the PTH chromatogram
after the N, just in that time they disappeared and the separation was nice
again. We have no idea what they are. The last week we got some PVDF blots, put
them on the sequencer and here the big peak (or should I call it mountain)is
back again. The amino acid peaks are detecable on this mountain, if the
sample was very concentrated, if not they are lost. The mountain comes up in 3
- 5 following chromatograms, after this there is nothing for x
chromatograms, then again for y....and so on. One idea is aniline, we checked and cleaned
the HPLC, replaced the tubings, cleaned the conversion flask, replaced most
of the PTFE tubings of the sequencer, new chemicals, checked for air in the
detector cell (negative, even during the mountain).
Strange that they came back after a blot (it was not nitrocellulose).
I am thankful for any suggestion.
