Peptide Acetylation

Hi,
I have a student who wants to acetylate internal lysine residues on
a peptide that represents the first 20 amino acids of histone H4. He has
read about using acetic anhydride in both aqueous and in pyridine/DMF
solvents, but has only the brief description of methods in papers, not the
detailed protocol he would like to have. He has the following questions,
which I am unable to answer. I assured him that there were experts out
there willing to help out, and I append his questions below.
