I haven't seen it mentioned in the literature for the past few months, but
I have used it myself. For peptides not containing Asp or Asn, or for the
part of a peptide before an Asp or Asn, I use 2% DBU/2% piperidine in NMP
(v/v). I make up a stock solution of 10% DBU/10% piperidine in NMP and
put this into the piperidine place on my ABI 430A. I raise the B regu-
lator pressure to deliver 1 ml of my mixture in the same amount of time
that 1 ml of piperidine would be delivered. This means I have to shorten
the amount of time for HBTU/HOBt delivery. At some future date, I'm
planning to set the B regulator back where it was, and to measure the time
for delivery of 1 ml of the mix, and adjust the RV cycles accordingly.
At any rate, the DBU/pip mixture has worked well for me, and I have been
able to couple successfully some residues that were considered "diffi-
cult".
