Hi all, PE (Perseptive) know we use DCM and have never suggested this
damages valves. Valve life seems to be variable, and generally short, no
matter what chemistry you use. I believe new valves are on the way.
Resins are problematic. All recent Gly resins we have used do not work.
A recent Ser batch appeared to shrink after first deblock, then strange
monitoring, closing the column space normalised the monitoring but the
synthesis failed. We got it to work by pre-swelling (vortex in DMF for
30min) then adjusting the column (15mm not 16mm even this may be important
below 0.1mmol for some resins!) so that there was no gap. Monitoring
normal.
Try Calbiochem-Novabiochem for resins.
