I will soon try to use a hydroxyapatite column (Tosohaas,TSK HA-1000, 5 =
=B5) for separation of glycoproteins and I have some basic Qs:
Are there other ways of elution than by Na (or K) phosphate gradients ?
I have seen a combinied negative CaCl2 (0.3 - 0.01 mM) gradient
and a positive phosphate gradient. How does Ca influence on the
elution? CaCl2 0.1 mM is often included to prolong the column
life time, I have understood.
