To all peptide synthesis specialists,
We would like to know if you could tell us honestly the failure rate over a
period of 3-4 months with the Pioneer w/out MPS and what were the causes if
known or suspected of these failures, i.e. valves, resins, columns.
The aim is not to complain or bully PE Biosystems but to let them know what
are the things that they need to address to make an excellent platform with
the same standards as their well known DNA sequencers.
Our chief technician with over 10 years of experience in peptide synthesis
has just passed on to me the following memo.
In it he includes data from the ABI 433.
Dec 01 98 to April 20 99
Pioneer
58 requests
39 made first attempt, 19 (33%) failed at least once on Pioneer. 11 were
successfully re-synthesised at 2nd or 3rd attempt. There was a total of 82
syntheses which included 24 repeats.

14 made first attempt, no failures, includes 8 which were unmakeable on
Pioneer.
This is favorable to the Pioneer as some syntheses were stopped very early
and the data discarded!!
Reasons for the failed synthesis are: resins and column diameter (16 vs. 15
mm)
