Hi Amanda,

Biotin will block the sequence if it is on the N-terminal of the protein,
however, if the N-terminal is not biotinylated, the protein will sequence
right through any biotin label that may be present without any problems.

You can get more information from the following :

Smith, J.S., Miller, B.T., Knock, S.L., and Kurosky, A. Biotinylated
peptides/proteins: I. Determination of stoichiometry of derivatization.
Analyt. Biochem. 197:247-253, 1991.

Smith, J.S., Miller, B.T., and Kurosky, A. Biotinylated peptides/proteins:
II. Identification of biotinylated lysyl phenylthiohydantoins. Analyt.
Biochem. 197:254-257, 1991.

Good luck,

Subject: ProtSeq: biotin-labelled proteins

Hi everyone,

A customer rang me to ask if biotin labelled proteins could be
sequenced.
I have no idea and so would appreciate your input. Can it be done? Does
anything special need to be done before starting sequencing?

Thanks in advance

Amanda Hall
