Dear Christer,
Last year we had a similar separation problem, all PTH-AA eluted
off earlier. Bob Apfelzweig from ABI told me that this problem occurs
when the column was stored for more than 6 months prior to installation on
the Procise, and he sent me a installation procedure which have been
working and preventing this phenomenon called "channeling" (new channels
on the column resin bed).
Starting equilibrate de column in high organic solvent
concentration (90-95% solv B at low flow rate 20-50ul/min for several
hours), and gradually increase the flow rate to 300ul/min, after this
deacrease in 10%B stepwise from 90% untill the initial conditions. The
entire procedure could take several hours.
