Dear all,
Glad the subject of silver staining turned up; I think I do have a problem
with the glutaraldehyde-free silver stain protocol. I recommend my
collaborators to use the -96 Shevchenko et al. protocol, and it almost
always works well, especially now that I employ the silver destain (F.
Gharahdahi et al Electrophoresis 20, 601-5, 1999) followed by nanocolumn
concentration/desalting. BUT, in a few cases when there were significant
silver bands I got absolutely no MALDI signal (just the trypsin autolytic
peptides). Has anyone noticed if there is any critical step in the silver
staining procedure? Somebody had experienced that too long sensitizing with
sodium thiosulphate screw up the gel. Anyone seen something similar? Anyone
has a fool proof silverstaining protocol to share?
