Dear DNA Sequencing Friends,

I'm interested in the reputed longer read protocols using TTE buffer
instead of TBE. Anyone out there using TTE? What's your verdict? When I
checked the cost of TAPS (the substitute for Boric Acid), I almost fell off
my chair. ABI's recipe for 1 liter of 10X TTE Buffer:

0.5M Tris-base 60.55 grams
0.5M TAPS-acid 121.65 grams (Sigma part number T9659)
10mM EDTA 3.72 grams

The cost (from the Sigma Catalog) of 1 liter's worth of TAPS is
approximately a whopping $40! ( 1kg - $355.50)

Is the stuff worth it? Are there less expensive sources?

Thanks for your help.
