Subject: ProtSeq-ABI477 & Porton
Dear All, My faithful 477 is misbehaving which is unusual,and I'm at a bit
of a loss for an obvious fix. So before I start tearing it apart, I'm
hoping someone can help narrow it down for me. Symptoms: Two different
protein stds, on pvdf and/or protein support, start off looking completely
normal for about three cycles, then the yield falls off and by cycle six I
don't get anymore sequence.
All I've done so far is leak check the R1 and R3, which appear to be fine.
Another problem on our Porton LF3000: Tyr is giving two peaks, one normal
in position and another eluting shortly after Ala. They are about equal in
height. Also Lys is giving two peaks ,normal (very small) and one before
Pro. I suspect these might be PTC's? What is the most likely fix
possibility here? Any and all help will be greatly appreciated!
