Dear Bill,
use a filter between your injector and your column. I use a M 560 precolumn
microfilter from Upchurch with a 0.5 um frit (A 735). This works very well
for 1mm and 300 um columns. I never filter any digest prior to injection
since the chances of material losses are far to great. I acidify the
digests to 1% TFA, sonicate the samples, give it a quick spin to get all
the solvent down and the inject the howl sample onto the column. If the
back pressure gets up change the frit. It is also good to use a filter
between the pump and the injector. Here I use the A 318 with a A 102 frit
also from Upchurch. This frit I have to replace more often the one on top
of the column!
