I am not knowledgeable about K12; however, we had excellent luck
with pTrc99 (Amann et al, Gene, 1987?) in a variety of strains, including
DH5alpha in expressing ras p21 (189 aa). In my limited experience, strains
do make a difference, but it has been hard for me to predict how.
