Hi Jeni!
In general Glycosylated Hyl does not seem to be washed out of the reaction
cartridge with the standard chemistry, as is found for other glycosylated
residues. However, when I sequenced a collagen some years ago I noticed a
double peak around the Tyr position (then working with an 473ABI) which
seemed to me to be associated with glycosylated Hyl. The reason for this
was that the position was always Y in the GLY-x-Y triplet sequence, that
the number of Hyl determined by AAA fitted to the sequence only when I
assumed that the cycles showing this double peak were counted as Hyl, and
most convincingly, there was a lysine which was apparently only partialy
hydroxylated and partially glycosylated so that I had in one cycle Lys, Hyl
and the double peak. (Eur.J.Biochem.210,839-847(1992),contains the
chromatogram).
Best regards
