Briefly, FTIR is usually used mainly to study the amide stretching
vibrations of the protein backbone, which are sensitive to hydrogen
bonding and therefore to protein secondary and tertiary structure.
Like circular dichroism (CD) it can measure the overall secondary
structure. However, FTIR is much more sensitive to beta structure than
is CD (for example it often can distinguish anti-parallel beta from
parallel), but less sensitive to alpha helix. Since CD is very good at
alpha helix but not so great at beta sheet, the two are highly
complementary. Like CD, FTIR can also be used to study thermal
unfolding of proteins.

One major advantage of FTIR spectroscopy is its ability to measure
protein conformation of solid forms of proteins such as lyophilized or
spray-dried powders, aggregates, precipitates, or films. This ability
has been exploited in pharmaceutical applications to optimize
stability and preservation of activity for lyophilized proteins, to
investigate mechanisms of aggregation, etc.
