Dina,
Thanks for all the info, but I still have a few questions. For the prep you
suggested a qiagen midi/maxi prep. Did you do any extra ethanol
precipitations that are not included in the protocol? Also in the post
reaction purification, have you ever used sodium acetate & 95% ethanol
followed by a 70% ethanol wash? This is the method I use for regular size
plasmids and it works well. I have used it on BAC and it works ok. There
still seems to be some left over uncorporated dye. I have run two BAC
samples that did work , they just looked a little "dirty" in comparison to
regular plasmid DNA. I an not sure if it is the way it was prepped or
something in the purification.
