Thiocarbamates are not very stable at low pH- remember that the
Edman degradation works because of acid-induced decomposition of a
PTC-peptide to an anilinothiazolinone amino acid, unstable also, which is
rearranged to PTH-AA. Of course, the cleavage is done at higher temp and
much stronger acid. Certainly the potential is there for decompoition.
This is one liability of the Waters "Picotag" system for analyzing
amino acids as PITC adducts- they decompose a few % a day at room temp.
under even neutral conditions in solution.
