Dear all,
we too have been using the destaining technique as described by the
Rockefeller group. Although it is difficult to quantify the yield, it is
absolutely clear that we get good MALDI spectra easier from low quantity
samples after the silver has been removed. The other day I analyzed about 2
pmol, loaded in two lanes, from a silverstained gel. One was destained; the
other treated directly and this direct comparison showed that destaining at
this level helps.
