Hey Jenny,
in my recent work it turned out theat i-propanole worked also well for
"bad" eluting peptides using standard columns. But after several test I found
that beside Vydac other C18s work well or even better, like the PPN material
from Macerey&Nagel, the Reprosil from a german company run by Dr. Maisch, or
the YMC material. Try first a propanole solvent and secondly change from
TFA to HCL (sorry I forgot the concentration, its about 10 mM) which reduces
the hydrophobicity of ion-pairing. You may also increase the temperature
during separation, we used our columns normaly at 35-37 C (dont smile about the
body temperature. But I agree that ion-exchange is a very good solution for
your problem.
