I started using this set a couple of months ago and noticed that the
larger multiplexes result in fainter signal--even the size standard is
quenched. I understand from the PE Bio tech support that loading too
much PCR product (salt) can quench the signal, but we see this effect
down to 1.5ul of product! With my own index marker set (FAM, HEX, TET)
and the TAMRA standard we could load up to 4ul of product and not see
any salt problems. Has anyone seen this and/or does anyone have any
ideas about why my problem seems so severe?
