We use a 5X sequencing buffer made by ABI (cat. #4305605 (600rxns) or
#4305603 (5400 rxns)). It's actually a very basic buffer (Tris 80mM,
MgCl2 2mM final concentration). We dilute the 5X buffer to 2.5X in
water and use 6ul buffer with 2ul BigDye. No difference in sequencing
quality noted yet and what an incredible cost savings!

Subject: Source for buffer

Dear colleagues,

I am interested in finding out where I could obtain buffer
(called Half Term
Buffer?) used to dilute ABI BigDye kits. Is there a commercial
source of
this buffer (in US, or better in Germany)? What is the
composition of this
buffer? Can be made in the lab?
Another question is regated to ET-terminator kits from Amersham.
Supposingly, these can also be diluted 4-fold. Is there any
experience with
this?

Thank you

Best regards Jan Ilavsky
