I have been having just a horrid time trying to ligate my gene into the
pKK223-3 vector from Amersham Pharmacia Biotech. I have tried different
T4 DNA ligase and different ligation temperatures, but to no avail.
Has anyone else had this problem? If so, how did you overcome it?
