Lisa,
I saw the group reply of alternate suppliers; that is OK, but you'll still
need to weigh out the reagents into clean bottles. That's a pain. I
remember from an APS meeting (Nashville?) a vendor sells preweighed kits for
these synthesizers; I don't remember who.

I've used TBTU on the 430 with less than stellar results. The metering loop
often clogged, most likely from precipitation. My experience is that
activators (uroniums) are on the edge of solubility and often clog /
crystallize in limited volume systmes like peptide synthesizers.
Substituting one for another is a risky proposition and one must be willing
to pay the consequences of that endeavour.

I've never tried PyBOP, as that is both expensive and unstable over long
periods of time. I did use something called ToPipu. It's a more stable
variety of TBTU. It worked very nicely and gave a stable solution with HOBt
for 6 weeks. I don't see it in the 1999 Novabiochem catalog anymore and
can't provide a supplier.

Hope this is helpful.

ABI is backordered on activator. Has anyone tried PyBop on the 430A or 433?

Thanks
