I had the same problem with low B-lactoglobulin yield. The low yield may be
due
to not be getting R2 delivery. Try manually opening valves 1,3,11,15. If
R2 is
not delivered to waste then the R2 valve (#3) may need to be replaced. R2
is
gas delivery and does not show up as a sensor error. I don't recall seeing
an
aniline peak and I have an explanation for that. Good luck.

Hello All and Merry Christmas,
Well merry for some of us. I'm in a delemia. Just recently my Procise Seq is
only producing 10% yields on B-lact standard run. I'm not sure why this is
occuring. There is also an ugly, extreemly large peak, which I've been told
is
analine, in the first residue cycle. This peak then drops off in further
residue
cycles. To reduce the aniline peak, I've done various washes with 70% ACN
0.1%
TFA, MQ, MeOH and finally S2B by using the cleanup menu. But I think the
aniline
and reduced yields are seperate issues.
>From what I've worked out, it can't be in the coupling or the cleavage
stages
b/c otherwise there would be carryover. This we are not seeing.
It can't be in the conversion b/c otherwise we'd see shadow peaks which we
aren't seeing.
Its not the column b/c the standards are fine and it must be in the flask or
chemistry b/c the blank is ok too. I recently changed the flask, optimised
the
post/pre conversion times and changed R2B to try and find a solution.
Anyone out there before the christmas period that can help with any
suggestions
would be much appreciated.
