I am looking for someone with extensive experience with the 3'-TAMRA CPG
from Glen Research. I have attempted to contact the technical services at GR
but they are closed for the holidays. Oligos were synthesized using the
3'-TAMRA support from Glen and the (tac)-amidites from Proligo. These
amidites boast a deprotection time of 15' at 55C in ammonium hydroxide (or 2
hours at RT). I would like to minimize the exposure of the TAMRA to ammonium
hydroxide. Will the 3'-TAMRA cleave efficiently enough in ammonium hydroxide
at 55C for 15' or will the (tac)-amidites be deprotected in 50 nM potassium
carbonate/methanol at RT? Any experience would be greatly appreciated.
